The MV Hondius is sailing towards Spain's Canary Islands where all passengers will be evacuated, after being anchored for three days near Cape Verde.
Three people have died either on board or after travelling on the ship after a hantavirus outbreak.
The cruise set off from Argentina a month ago. The cruise ship will dock at Granadilla port in Tenerife, Spain's health minister Monica Garcia said on Wednesday.
Everyone on board will undergo a medical assessment on arrival and, if fit to travel, those from abroad will be repatriated to their home countries, Garcia said.
People in Tenerife told the BBC that they are concerned about the ship's arrival and that residents "could end up with a problem on the island".
"I don't think it's a good idea... but people do need help", said one of the residents.
