The host of Would I Lie to You and Destination X will make his return to acting in six-part show Bill's Included.
The show will follow Brydon's character as he adjusts to an unconventional living arrangement with a group of students.
Brydon said he felt "very lucky" to be part of the project, which has started filming.
The new series will follow Brydon's character Bill Bream who is forced to rent out his spare rooms following his divorce.
It follows him finding his feet as a landlord and housemate to Gen Z lodgers.
Brydon said: "I feel very lucky to be part of Bill's Included. Alex Owen and Ben Ashenden have written some truly wonderful scripts and I can't wait to help bring them to life."
The show will also star Dylan Brady, Ada Player, Harry Baxendale and Yasmine Al-Khudhairi as the students who become part of the unlikely house share.

Brydon is also one of the series' executive producers, alongside Alan Partridge star Steve Coogan.

Brydon is best known for his portrayal of Uncle Bryn in Gavin and Stacey which was set in Barry.
He also lent his voice to the Christmas Day adaptation of Julia Donaldson's book, Tiddler, and recently became a published children's author.
